Homeobox protein CDX-2 is a protein that in humans is encoded by the CDX2 gene. This gene is a member of the caudal-related homeobox transcription factor family that is expressed in the nuclei of intestinal epithelial cells. CDX2 is also used in diagnostic surgical pathology as a marker for gastrointestinal differentiation, especially colorectal. Function: Involved in the transcriptional regulation of multiple genes expressed in the intestinal epithelium. Important in broad range of functions from early differentiation to maintenance of the intestinal epithelial lining of both the small and large intestine. Subcellular Location: Nucleus. Post-translational modifications: Phosphorylation of Ser-60 mediates the transactivation capacity. Similarity: Belongs to the Caudal homeobox family. Contains 1 homeobox DNA-binding domain. Paraformaldehyde-fixed, paraffin embedded (Rat colon); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (CDX2) Monoclonal Antibody, Unconjugated (ascites of bsm-33063M 5G8) at 1:2000 overnight at 4°C, followed by a conjugated secondary (sp-0023) for 20 minutes and DAB staining.
