EIF5, Eukaryotic initiation factor 5, catalyzes the hydrolysis of GTP bound to the 40S ribosomal initiation complex with the subsequent joining of a 60S ribosomal subunit resulting in the release of eIF2 and the guanine nucleotide. The subsequent joining of a 60S ribosomal subunit results in the formation of a functional 80S initiation complex. Function: Catalyzes the hydrolysis of GTP bound to the 40S ribosomal initiation complex (40S.mRNA.Met-tRNA[F].eIF-2.GTP) with the subsequent joining of a 60S ribosomal subunit resulting in the release of eIF-2 and the guanine nucleotide. The subsequent joining of a 60S ribosomal subunit results in the formation of a functional 80S initiation complex (80S.mRNA.Met-tRNA[F]). Similarity: Belongs to the eIF-2-beta/eIF-5 family. 25 ug total protein per lane of various lysates (see on figure) probed with EIF5 polyclonal antibody, unconjugated (bs-10035R) at 1:1000 dilution and 4°C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
Paraformaldehyde-fixed, paraffin embedded(rat pancreas); Antigen retrieval by microwave in sodium citrate buffer (pH6.0) ; Block endogenous peroxidase by 3% hydrogen peroxide for 30 minutes; Blocking buffer (3% BSA) at RT for 30min; Antibody incubation with (EIF5) Polyclonal Antibody, Unconjugated (bs-10035R) at 1:400 overnight at 4℃, followed by conjugation to the secondary antibody (labeled with HRP)and DAB staining.
